How a K.O Works
The goal is to repace a WT gene/gene of interest with a selectable marker (SM). This SM is on a plasmid and will have primers and homology arms (H.As) added to it to create the K.O PCR SM fragment. During homologous directed recombination (HDR), the K.O occurs!
The Knock-in process
Now that the SM is in the endogenous location of the WT gene and we've confirmed the K.O, we can K.I! The same process will occur. A plasmid with the gene/mutant of interest(G/MOI) undergoes PCR with primers and H.As. This amplicon is then added in during transformation. Goal: GOI/MOI knocks into place!
