Lab _ practical 6Observation of plant epidermis

START

Objective:

The aim of this practical is to observe epidermal cells of different species trying to discover their main structures and to establish comparisons between them.

Materials used:

  • Microscope
  • Onion bulb & leek
  • Leave
  • Slips and cover slips
  • Lab coat
  • Forceps
  • Stirring rod
  • Nail polish (transparent)
  • Celo tape and scissors
  • Water

Basis:

  • Research information on the structure and function of the plant epidermis.
  • Find information on the structure and function of stomata.

Methodology of the onion and leek.

Later on, we put the sample in the slide and we add a drop of water and we cover it with the cover slip.

We take the eopidermis of the inside of an onion, carefully trying to take the thiner layer as possible.

Lastly we put it under the microscope and we observe the onion. We then repeat the same process with the leek.

Later we take a bit of celotape and we put it on top of the nail polish that must be completely dry and we take it off the leave carefully so that we can see something under the microscope.

Once we have the sample we put it in the slip in a way that the celo sticks to it so you don't have to put any coverslip. Lastly we observe it.

In order to try to observe the epidermis of a leave we have to do a different process. Firstly we paint the bottom of the leave with transparent nail polish and we let it dry.

Observations from the onion an the leek:

  • After observing the epidermis of the onion under the microscope we couldn't find stomata as it is a simple epicelyum that has no contact with the light so it was not able to perform photosynthesis.

Observations from the leave:

  • Lastly, when we saw the leave we could observe the stomata as the leave was able to perform photosynthesis. We watched the bottom part of it because if the stomata was located in the top of the leave the water would evaporate.
