UV- Visible Spectroscopy follows the Beer-Lambert Law
Arnold Beckman, the founder of his eponymous company, introduced the DU UV-vis spectrophotometer in July 1941. It was the official version of the Model D prototype that he and Howard Cary had created.
Sample Handling Techniques
Basic Instrumentation
- Uv-visible spectra are usually recorded either in very dilute solutions or in the vapor phase.
- THe sample is dissolved in some suitable solvent which does not itself absorb radiation in the region under investigation.
- Commonly used solvents are cyclohexane, 1,4-dioxane, water and 95% ethanol.
- The chosen solvent should be inert to the sample.
There are 2 main types of spectrophotometer used to measure the absorption of U.V. or visible radiation: 1. Single beam UV-Vis. spectrophotometer 2.Double-beam UV-Vis. spectrophotometer
Instruments for measuring the absorption of U.V. or visible radiation are made up of the following components;
- Sources (UV and visible)
- Wavelength selector (monochromator)
- Sample containers
- Detector
- Signal processor and readout
